Salmonella Petri Film Test instructions

First, the principle

The test plate contains a dry powder medium, a selective inhibitor, a chromogenic substrate, and a cold water soluble gel. The medium provides nutrients for the growth of Salmonella; selective inhibitors can effectively inhibit the growth of other bacteria; when Salmonella grows and develops, the chromogenic substrate is decomposed and frees the chromogenic group, and the Salmonella colonies are black, brown or gray.

Hydration test piece

Place the test piece on the surface of the water platform, uncover the upper film, suck 1mL sample and add the diluted droplets to the culture layer, slowly cover the upper layer film, align the grooved part of the liquid plate to the sample dilution area and place it on the upper layer film. The finger presses the liquid plate to spread the diluent evenly in the pressure ring. 2 repetitions per dilution.

Streaking

The upper layer film is uncovered, and the inoculating ring is used to pick up the Salmonella enrichment liquid to draw a line (continuous zigzag line) on the upper part of the plastic film and the hydration culture area of the culture layer, and the upper layer film is covered in the hydrated culture area.


Cultivation

The inoculated test pieces were stacked (not more than 15 pieces) in a Ziploc pocket (not sealed), cultured at 36 °C ± 1 °C for 24 h ± 2 h, and the negative result culture time was extended to 48 hours. The test piece should be placed in a flat place (a small piece of cardboard can be placed in the incubator), and it should not be placed directly on the wire frame to facilitate the deformation of the gel.

Interpretation

After the end of the culture, the test piece was taken out, and the black, brown, and gray (regardless of the color depth) were Salmonella. The waste is treated by autoclaving at 121 ° C for 20 minutes.

Biochemical identification

The upper film of the test plate was uncovered, and the suspected positive colonies were aseptically picked for biochemical and serological identification, and the method was the same as GB 4789.4.

Fourth, storage

Store in a cool, dark place with a shelf life of 1 year; store in a self-sealing pocket at room temperature after opening, and use up within 1 month.
