First, the principle

The medium consists of nutrients, selective inhibitors, indicators and cold water soluble gels. Nutrient provides the nutrients needed for the growth of coliform/heat-resistant coliforms; selective inhibitors can effectively inhibit the growth of other bacteria; colony formation, the indicator is red; cold water soluble gel can effectively trap the coliforms to break down lactose the bubbles produced. The dry powder medium is sterilized by radiation and does not need to be heated and sterilized.

Vaccination

The dry powder medium was torn open in a sterile manner, poured into a 500 mL sterile flask, and shaken to uniformly distribute the dry powder medium at the bottom of the flask. Pour 100mL of water into a triangular flask and shake quickly (shake for 3s-5s, do not shake too much to avoid large bubbles in the gel; when the solution becomes viscous, do not shake), let stand for 20min to make the gel completely solidified. If the water sample is highly contaminated, dilute the water sample (take 10mL water sample, add 90mL sterile water to mix), and select the appropriate dilution for testing.


2. Cultivation

The total coliforms were detected in the incubator at 36 °C ± 1 °C for 24 h ± 2 h; the heat-resistant coliforms were detected in a 44.5 ° C ± 0.5 ° C water bath for 24 h ± 2 h.

3. Count

After the completion of the culture, the flask was taken out, and the red dot-folded colony was coliform/heat-resistant coliform, and the unit was CFU/100 mL. If the whole medium turns yellow, there are many red spots and fine bubbles. The waste is treated by autoclaving at 121 ° C for 20 minutes.
